New membrane material containing macrocyclic compounds for affinity adsorption of biomolecules |

Abstract: |
Using polyether sulfone(PES) and ethylene-vinyl alcohol copolymer(EVAL) as membrane matrix material, cyclodextrin macrocyclic compounds was adopted as functive, the reactive dyes ligands were coupled to the basement membrane by making use of the rich active groups on the functive, and a new type of affinity membrane containing cyclodextrin was prepared. The effects of the CD content and, the pH of BSA buffer solutions, ionic strength, solution concentration and adsorption time were investigated. It showed that the optimum adsorption conditions are; pH:5.0, cyclodextrin content:10%, ionic strength:0, solution concentration:1.0g/L and adsorption time:4h, adsorption capacity reached 57.7mg/g under the optimum adsorption conditions. The unique cavity structure and properties of cyclodextrins(CD) with hydrophilic external and hydrophobic internal cavities are presented to induct and pack biological macromolecules such as protein. The existence of cyclodextrin macrocyclic compounds benefits to the performance of the membrane adsorption. The adsorption capacity was enhanced with the increase of CD content in the membrane adsorbents within limits. |
