抗氧化聚砜血液透析膜的构建与性能研究

摘要:
 为了抑制血液透析过程中氧化应激现象,以聚砜 (PSF) 为膜材料,水飞蓟宾-卵磷脂 (SPC)为改性剂,采用浸没沉淀相转化法制备抗氧化的SPC/PSF血液透析膜。随着SPC在铸膜液中含量提高,SPC/PSF杂化膜纯水通量增大到187.9±6.9 L/ (m2h)、亲水性提高,SPC/PSF膜对1,1-二苯基-2-三硝基苯肼(DPPH)和2'-联氨-双-3-乙基苯并噻唑啉-6-磺酸(ABTS)最高清除率可达到88.3±1.3%和99.4±0.3%,抗氧化性提高。SPC/PSF膜相比于纯PSF膜不仅血小板粘附量减少,细胞增殖能力增强,而且具有较强的抑制超氧化物歧化酶 (SOD) 产生的能力。结果表明该SPC/PSF血液透析膜可望应用于血液透析领域,缓解病人的氧化应激状态。
 The objective of this study is to improve the antioxidant properties of polysulfone (PSF) hemodialysis membranes through hybrid silybin-phosphatidylcholine complex (SPC). The SPC/PSF hemodialysis membrane with antioxidant activity was made using PSF as membrane material by liquid-liquid phase separation method in the presence of SPC as modifier to suppress hemodialysis induced oxidative stress. For the SPC/PSF hybrid membrane,the pure water flux was increased to 187.9 ± 6.9 L / (m2h) compared with the pure PSF membrane. With the increase of the content of SPC on the membrane surface, the antioxidant properties (the ability of scavenging DPPH and ABTS free radical) were increased to 88.3 ± 1.3% and 99.4 ± 0.3%. For the SPC/PSF hybrid membrane, the stability of free radical scavenging capability under typical hemodialysis condition was good. Compared with pure PSF membrane, the SPC/PSF hybrid membrane had good cell compatibility. Compared with the pure PSF membrane, the platelet adhesion of SPC/PSF hybrid membrane was decreased, and the blood compatibility was improved. The SPC/PSF hybrid membrane had strong ability to inhibit superoxide dismutase (SOD), while pure PSF membrane stimulated the production of SOD. The results indicated that hybrid modification by SPC and PSF provided practical application of the membranes with good antioxidant properties; and a new type of SPC/PSF hemodialysis membrane was expected to be applied in the field of hemodialysis and alleviate the oxidative stress status of the patients.
